I have Polar, hydrophobic compounds, the compound is combination of antibiotic and thiodizine group. it dissolve very well in DMSO ( 0.5 molar). But the problem is when i further dilute it (2ul/10ml like 100uM) in media it precipitated. i tried lower concentrations as well and different solvents like methanol, PBS, media (not soluble in them) combination of these solvents with DMSo, Nothing works. when i add my culture in plate which also consist media it precipitates my compound. and i can not change these media because it is the main media for parasite growth.
kindly help me how to avoid precipitation of my compound in media.
hello huma, have you filtered your compound after dissolving, if not then first filter with suitable membrane,
If the anti biotic is polar ,it is possible to use water or mixture of different ratio of water and dmso to obtain the maximum solubility profile .
while diluting the DMSO-drug solution by media, don't add your media to DMSO solution directly, it will precipitate your drug. Instead of that, add your DMSO-drug solution to media slowly (ie; add 2ul DMSO solution to 10ml media). This may not give a precipitate.
