I need paper of comparison of the effects of different cryoprotectants on mesenchymal stem cells and peripheral mononuclear cells. It would be better if its about Dextran-40 and HES.
My experience and research is on PBMC/DC more than Stems, but I would caution against using starches such as Dex-40 alone to protect cells during cryo:
1. you will lose cells during dehydration/re-freezing due to intracellular and extracellular ice crystal formation. Dex-40 can help with the extra-, but you need something else
2. DMSO, while toxic, is still the go-to as it is polar, replaces water in the cell, and is minimally toxic at 5-10%
3. Think about a mix of dex-40, DMSO 10%, and Human Serum albumin to make the cells feel like they're at home. Think about preserving cells in all dimensions.
4. Trehalose sugar is useful for preventing ice crystallization
