We are using the Endogro-SCME004 medium for cultivation of hCMEC/D3 cells, but since it is out of stock for a while, we decided to switch to Lonza. However, we just realized that there are stark differences between the media supplements b/w both the kits.
In short:
the Endogro is the most widely used and consists of LS supplement, EGF, Glutamine, Hydocortisone, Heparin, Ascorbic Acid and FBS. We also add FGF to it. Lonza EBM-2 comes with FBS, Hydrocortisone, hFGF, VEGF, iGF, Ascorbic acid,hEGF, GA-1000, Heparin. I was thinking whether all of these are essential for BBB monolayer formation... for example, VEGF is known to disrupt the BBB.
What components could we "skip" from the Lonza Kit, to keep things similar b.w the Lonza and the Endogro medium, to ensure good formation of a barrier?
