1. Can I assume that it is better to use 5 parameter logistic (5PL) fit than the 4PL for sigmoidal shape curves?

(4PL assumes symmetry of the standard curve around the inflection point where the 5PL does not - and in case of immunoassays, I think that curves are more often asymmetrical; moreover just in case of the symmetry equation for 5-PL equals 4-PL.... so 5-PL may SEEM to be more universal).

2. Shall I use weighting with 4- or 5-PL curve fit? If yes, which would be better 1/Y or 1/Y^2?

3. Is it possible to have universal protocol for fitting to the standard curve (of sigmoidal shape) in ELISA tests i.e. "5-PL with 1/Y^2"? or it would be better to check the fit every time?
