The point is that I have to establish an ex vivo system of lymphoma cells from human patient because I want to see the effect of an immunotherapeutic antibody in terms of the following :

1) The response of the malignant cells.

2) Optimizing the right dose.

3) Trying to evaluate the T cell biology especially in the resistant or relapsed samples.

So the cell lines won't give me the chance to mimic the physiology.

It is possible to culture and expand primary lymphoma cells isolated from patient material. I have tried this with 3 different (DLBCL) patient samples and accomplished this with 1, so 1/3 succes rate so far. We cultured the lymphoma cells on a monolayer of irradiated mouse fibroblasts as a source for CD40L with medium (IMDM, 10%FBS) supplied with IL-4. As an alternative for CD40L can maybe try to order and just supply this in your medium, but irradiated mouse fibroblasts also do the trick.

I had to be very patient but after 3 months I have expended a sample of 3x10^6 cells to 12x10^6 cells. Of course it is diffucult to say whether these cells are representative of the original patient material, to be sure it might be wise to check the STR profile and maybe some known hotpot mutations, but as far as I know it is as close as you can get.

Regarding your question, I am investigating the effect of Bi-Specific T cell engager antibodies "BiTEs" on lymphoma patient samples where we either consider the residual autologous T cells within the sample or use allogeneic T cells.
