I have been trying to culture P. falciparum, but I see contamination after ~1 week of culturing. What could this organism possibly be (image attached)? The culture media consists of RPMI, HEPES, sodium bicarbonate, Albumax, human serum and gentamicin.
The images in the original post: contaminants observed in P. falciparum culture stained with 10% Giemsa.
I decided to maintain human RBC in culture media without parasites to make sure that the contaminant is not coming from the parasite stock.
On the 3rd day, I observed specked particles.
On the 7th and 8th days, I started observing ring-form particles. These particles resemble the contaminants seen in the parasite culture, but without the parasites, they were smaller and had a lower intensity of staining dye.
Day 7 and day 8 looks like some candida
Is the giemsa fresh ? Filtered ? It's not artifacts from the giemsa ?
