I am carrying out an adherent cell culture of mcf 7 cells, they are currently fixed and therefore I can't use the protocol i would normally use for cell counting (hemocytometer). I was therefore wondering if anyone has any advice on how to carry out a cell count using a graticule?

I never used MCF-7, but with our cell line (PC12) we used a rubber scraper to detach the cells from the bottom, homogenized the suspension with a papeur pipette and then counted the cells with a Malassez chamber, we usually got good results. If your cells are tightly adherent you may either fixed them with methanol and then count the nuclei under a microscope using a specific coloration. Another way is to measure the amount of DNA present in your vials, there exist plenty of protocols described to determine DNA using either fluorescent methods or colorimetry.
