I am a little confuse what the condition of cells when I add a kind of protein the cancer cell uptake glucose, but the cells upregulate the apoptosis associated protein and the capacity of migration was inhibited. Could cells uptake glucose trigger some key factor might induce cells death?
To answer your question it is necessary to know more details:
What kind of protein do you add? What media you use? How much glucose is in the media?
Usually cancer cells have an increased uptake of glucose which is 10 to 17 folds the uptake of normal cells.
So that the increased uptake of glucose is not the reason for the apoptosis.
@Tomas Koltai, professor. We use a small molecular peptide granulin in our experiment. The hepg2 cells were assayed for glycolysis metabolism in the 4.5g DMED medium. Compared the control without peptide and granulin gradient concentration, the hepg2 cells with peptide was uptake glucose in the individual cell, but the migration abilities of cells were also inhibited.

When you used the peptide the glucose uptake increased or decreased?
Normally hepg2 cells have a high glucose uptake, even without any glucose uptake stimulus.
If this peptide used in the experiment increased even more glucose uptake, that means that probably a very higy load of lactic acid may have been generated in the cell. This should lead to apoptosis if the cell is unable to handle the excess of lactic acid (which depends on monocarboxylate transporters 1 and 4).
May be an interesting experiment should be to measure lactic acid production with and without the peptide.

If the glucose uptake is increased and ATP production and lactic acid are not increased, this means that the cell is not increasing aerobic glycolysis (Warburg effect) nor OXPHOS.
The only situation that would explain this conduct is that the metabolism shifted towards the PPP (pentose phosphorylation pathway). This usually happens when the cell is in a great need to generate NADH.
