One day after the last split of my MC3T3-E1 culture (P23, L-MEM+ 10 % FBS+ Pen/Strep) these big particles were turning up in the cell culture flask. I can't remove them. Is this a contamination?
Hello Maria. I will say, no, it is not a contamination. If you look at it carefully, the focus is either on the cells or the big particle. When the cells are on focus, the big particle is blurred, and this happens vice versa. This means, that the big particle is sticking on the other side of the plasticware - most probably within the plastic itself (artefact from the manufacturer), or outside the plastic (got sticked on the surface).
Culture your media (L-MEM) and FBS alone and in combination in 6 well plate and observe for 72 hours. Also culture PBS which you are using. Contamination of this type is most probably because of FBS, change FBS.
Looks no, if you add FBS in culture media without centrifuge, these type of particles can be seen.
observe/incubate for next 24 hours to see if it is growing.
