Table for some medium used in differentiation of Mesenchymal stem cells adipocytes tissue
With some cultures it happens quite efficiently without any induction at highish confluency - it can be a nuisance when trying to expand.
to deliberately induce:
grow to confluency. change to induction medium:
DMEM + glutamine, 10% FBS; 1uM dexamethasome; 0.5mM IBMX, 10ug/ml insulin.
leave for 2-3 days. change to maintentance medium:
DMEM+glutamine+10% FBS + 10ug/ml insulin. leave for 2-3 days
repeat so that induction has had three rounds. On the third maintenance, leave for 7-10 days. or stop whenever you are happy with the progress, it is quite obvious under the microscope.
stain with oil red o to satisfy science.
All the protocols available can convert mesenchymal stem cells into white adipocytes.There are no protocols available for converting into brown adipocytes.One has device on their own.In fact it will be a million dollar discovery.
