@Lazaro Molina Sir this HCl I am going to use for cell culture use (Just to maintain pH of media ). this is the reason I want sterile 1 N HCl.. if it is not autoclaveable , so please give me any alternative for this ..
Make the medium with buffer phosphate, better to maintain the pH of the culture… Or add the HCl to the culture medium (the quantity normally is low, then you can autoclave the medium with the appropiate pH. The best solution is buffer phosphate
Have you tried with a 0,22um filter? I do not know if it is possible to filter at that concentration but you can ask for a special filters to do it.
Other thing you can do is to add the HCl to the culture medium and filter it. I had a problem with DMSO because it can not be filtered with normal filter. Therefore, I added DMSO to the culture medium at 0,5% and the solution was filtered.
In case that precise concentration of HCl is very important for you, I recommend you sterilization through filtering, because the concentration concentration may be reduced during the procedure of autoclavation. If you want to use 1N HCl only to pH change in culture media, this solution is autoclavable.
as mentioned above, you can use double distilled water to prepare culture medium, and use filters to sterilize the HCl ( prepared with DD water ), anyway you are going to autoclave the medium after PH adjustment, , ,
Autoclaving HCl will decrease the concentration since HCl will get into the gas phase when heated. Use autoclaved water and dilute the amount of HCl you need from concentrated HCl. But I actually doubt that this is necessary.
no need to autoclave just add it to the medium after autoclaving it under aseptic conditions. if you want to use it in adjusting the pH , add it or NaOH to the medium before cooking the cooke and autoclave.
@ Essam Kotb ...Sir, cell culture media contains lot of proteins and growth factors, If we will do autoclave these contents will degrade ??? If I am wrong so plz tell me ...& plz send me any reference for this..
Actually Media contain various amino acids and co-factors which will degrade when undergoes autoclaving. just adjust pH of media and then filter in aseptic condition.
