We require a method that will keep the sample in tact while removing it from Wax embedding, so that it can then be embedded in EPON in order to avoid damage to the instrument during scanning electron microscopy.
I assume you mean paraffin wax;
-remove the excess wax from the block with a razor blade
-soak the sample with excess amounts of fresh xylene for 1 day
-replace the old xylene with new xylene every day for 3-5 days depending on how big your sample is in the block
-wash the xylene out of the block in multiple distilled water washes
-treat the material like fresh tissue; dehydrate and process as you normally would for embedding material into Epon.
Why embed material into Epon in order to avoid damage during SEM ?
