Dear colleagues,

I am studying microglia in early development. I would like to design a study where I deplete microglia in neonatal pups using a CSF-1R inhibitor.

Several studies that employ the strategy of ablating microglia deliver these drugs via chow. However, neonatal pups cannot yet feed themselves, and there is no published evidence that sufficient traces of the drug can be passed via the maternal breast milk to deplete microglia in the pups.

Has anyone tried administering CSF-1R inhibitors via IP? If so, are there any specific protocols to prepare the drug for delivery? Does it need to be diluted or mixed in PBS or DMSO for example? How do you determine the appropriate dose to inject? How often should injections be administered?

I appreciate any helpful insights in advance.

Best,

Alicia
