Hi, I'm looking for a lysisbuffer that can make the proteins from oats available for a Bradford protein assay. RIPA and SDS are both compatible with the kit I'm planning to use (thermofisher) and I'm wondering whether either of the two buffers can be used for making the proteins available for analysis and if they both work, which one is better for oats. As far as I know, it doesn't matter if the protein gets denatured with a Bradford protein assay.
You may use both and extract can be used at Bradfrod measurement after proper dilution has been applied. RIPA is mostly the preferred lysis buffer and already includes SDS. You may add SDS and increase the content if it is needed. Do not forget to homogenize the sample and definitely apply physical disruption techniques to efficiently get inside the cells of the oats.

You should well pulverize and make a powder (texture like oat flour)..but this is just a simple and first step of the extraction encompasses the preparation of the sample prior to extraction. By defining the physical disruption I mean conditions like cell lysis...For this combine the RIPA and ultrasound and/or beat beater treatment to fully solubilize the oat proteome...
