I am working on Pseudomonas fluorescent strain identification, I found that some of isolate that I have been conserve at 80c last year have loosing their fluorscent appearance on new culture. someoine can explain the cause?
It is commonly seen, if we preserve any of the microbial culture for long time, they become functionally inactive, and take a good amount of time to regain their functional and metabolic potential back. Same thing happened with your Pseudomonas culture, please revive your culture again and again, it will definitely work.
Yes, Pseudomonas aeruginosa can lose its fluorescence under UV, if the cultures are left at room temperature for a short time. Fluorescence reappears when plates are re-incubated.
