We are cultivating keratinocyte. However, when the subculture is carried out, the shape of the keratinocyte becomes elongated. Keratinocyte was cultured in the same manner as before the subculture. If someone observes this shape when cultivating keratinocy, please advise.

I usually cultivate human primary keratinocytes up to 3-4 passages after their isolation from epidermis. Then, cells progressively switch from a polygonal shape to an elongated shape (like fibroblasts) and i avoid using these cells for my experiments.
The culture of Keratinocyte cells without feeder cells becomes elongated like fibroblast after some passages. but if you are using feeder cells for the culturing of keratinocyte than the shape of keratinocyte does not change even after passaging.

In fungi, morphological changes occur via mating hormone (please see file; tremella 2D-EP). Then, feeder cells may consume putative differentiating hormones produced by Keratinocyte.
