It is described in the literature that Y27632 proliferates cells and inhibits differentiation. Cell adhesion is also good. But when we experimented, there was no clear difference. I was experimenting with keratinocytes and used serum-free keratinocyte medium. Please let me know, if anyone have experience using this material.
In a different lab, I used it to inhibit rho kinase when treating cells with Rho GTPase activators. I found inhibition was unpredictable (maybe lot dependent?). I never noticed effects on proliferation.
I have used Y27632 (Stem Cell Tech) to induce embryoid bodies from iPSCs and I have to say that it worked perfectly at a 10uM concentration (in fact, without it there was no cell adhesion at all). Regarding cell proliferation, I used it at same concentration as for cell adhesion but I didn´t see any clear difference.
I have used Y27632 for enzymatic method of iPSCs subculture process. It really helps to enhanced the cell survival and adhesion without affecting the stem cell properties.
As far as I know, this compound is not quite stable, so maybe should be more careful about the storage of it.
