When keratinocytes lacking maternal cells are cultured, does keratinocyte damage occur by treating mitomycin C(4ug/ml) and cholera toxin(10^-10M)? Or inhibit the growth of keratinocyte?
Hi: I am not sure your concentration of mitomycin C damages keratinocyte. But we have used mitomycin to damage the chromosome and inhibit cell growth. This is useful if we were to treated the feeder cells with mitomycin for the purpose of growing stem cells ; the feeder cells continue to produce growth factors, nutrients, etc. for the stem cells but they don't divide. Cholera toxin often acting as an agonist that stimulate the cells via receptor on the plasma membrane.
