I want to do a fermentation test for my yeast. I want to know if i can do it using YPD broth supplemented with phenol red as pH indicator. If can, how much phenol red would it needed, and how to formulate the broth as a whole. It would be very helpful if you attach some literature. Thankyou.

To investigate fermentation patterns of the yeast, first eliminate the carbon source from the medium and than add carbon source of interest to the medium at a specific concentration. There are two different things carbon assimilation assays and carbon fermentation assays. Later one can be noted by presence of gas in durham tube. Change in the pH and production of the gas are the preliminary measures to determine carbon fermentation pattern of micro-organism. Well I'm not sure about the use of congo-red but bromothymol blue indicator could be used for the same.
