I am trying to run a TaqMan assay where my housekeeping probe is labelled with VIC. I do not have access to a qPCR machine with VIC specifically defined, but HEX is recommended as a substitute. Is it faesible to set the machine to HEX and use the VIC probe anyway? Do the wavelength values need to match exactly?

HEX is definitely close enough for your needs. I've used HEX for VIC dyes before, in multiplexed assays with FAM dyes, and have had zero problems. Further multi-plexing with more dyes, however, might be deleterious.

You can definitely use it since these dyes have the closest wave length of emission and excitation. VIC has excitation of 538 vs 535 nm for HEX. They are far from other dyes like FAM which is 495 nm.
