Nowadays, we can find kits for total RNA extraction and miRNA extraction. I would like to know whether it is good to use total RNA to run 2 steps RT-PCR for miRNA expression. I will take total RNA to convert cDNA first followed by qPCR. Will this way be sensitive enough to detect miRNA expression?

I found that some recommended that we have to use miRNA extraction kit only and use this miRNA as template to convert cDNA rather than from total RNA.

I am confused. Which one will be a great way to go and shows sensitive enough? Thanks
