Hello Everyone.
I have samples that significantly vary in protein concentrations. can I load equal volumes of samples that have different protein concentrations on basis that later on I will normalize the target protein to the housekeeping protein?
Please I would be grateful if you advise.

mm you possibly could but just a heads up that your samples with high protein content, if they are too high (of a concentration), depending on your detection method, you may approach the detection limit in those lanes and you may not be able to accurately obtain proper loading controls/normalizations
It would be much better to adjust the loading volumes in order to load at least roughly the same amount of protein per lane. As mentioned by Aaron, this is important in terms of detection limits and proper normalization.
