Organoid culture is now well established method of growing goblet cells. It was said that these mucous secretory cells after isolation from the tissue die when put on culture plate. So is there any means of culturing them apart from employment of this pricey technique?
Hi Rohit, as far as I know there is not a method to isolate or maintain a pure goblet population. Using Ulex (aka UEA) will certainly enrich for secretory cells if you have access to flow sorting or a magnetic separation apparatus. Some of the researchers I know have started using Intesticult media from Stem Cell Technologies because it is much cheaper than adding the recombinant growth factors (EGF, Rspo, Noggin) used in the traditional "Sato Culture" and they see good growth. The downside is that you don't get to know exactly what is in the media.
@Richard: Exactly Sir. I was successful in isolating them from colon of mice but unable to grow them as they are rapidly dying. I am aware of the intesticult but the drawback is it is extremely expensive. I was successful in identifying pure goblet but proving via IF, Western.

In the small intestinal organoid culture the Goblet cells are present, and you can detect them e.g. by the PAS staining: http://www.hindawi.com/journals/sci/2016/3710836/; I hope this information will help you with the colon-derived Goblet cells.In case of IntestiCult medium, you need to take into account that this medium is designed for the small intestinal organoids; for the colon organoids it is necessary to add additional growth factors e.g. Wnt3a (http://www.ncbi.nlm.nih.gov/pubmed/22406745).
