If suppose the drug concentration is 10nM which is to be added, how do we calculate the amount to be added as final volume in 1mL of cell medium?

Dear Rohit,
10 nM = 10ng x MW/1000 mL
= 1 x 10 ng x MW/1000 = 0.01 ng x MW/mL (unit is ng/mL).
Therefore, you should weigh 0.01 x MW (nano grams), dissolve in cell medium and add sufficient volume of the medium up to 1 mL (total volume 1 mL).
