Dear All,
I surprised when i saw Agrobacterium tumefaciens colonies streaked on YEM agar plates continue to grow even after i stored at 4degree celsius. After 1 month the colonies overgrow and finally lose viability. I don't know how it happens. The fridge is working good without any fault.Can anybody tell me what and where the fault is and the reason behind this? Thanks in advance.
Agrobacterium indeed can grow (very slow) on plates in the fridge. I stored plates (LB medium) up to two months before transfer of colonies to fresh plates.
You should check about any technical issue during the storage period. generally bacteria do not grow at freezing temperature and remains in viable form up to an extent.
yes but this depend to situation of storage and the time length
