Even at 0.1% of DMSO could be defined that it slightly decreases cellular functions according to some authors, and this was demonstrated that it gets significant at 0.5%. Use as low as possible DMSO is the best practice and you should always have a solvent control. But I must still add that type of the cell you are using would always make difference for sure. I agree with Kaan. DMSO is cytotoxic at higher concentration. It may dissolve the cell membrane. At 0.5 % and above you may lose your cell. Maximum 0.1% is highly recommended for all type of cells.
I have successfully used up to 1% DMSO for these cell lines (and many other colorectal lines) - some cell lines can be sensitive to DMSO, sure (certain breast lines are quite sensitive). As long as your untreated control has 1% also (which you normalise cell survival to, of course) as well as consistent 1% DMSO for all your dose points you will be fine. It is a trade off regarding your compounds solubility in aqueous media versus keeping your cells happy...
hi - I can't say I have seen it published specifically [although our group will be soon]... We've just been doing it routintely for the last five years, where we have tested over 2000 compounds across many colorectal lines, including SW480/SW620.
