Basically, I am working on two yeast strains: Kluyveromyces marxianus K-21 and Pichia stipitis 21777. K-21 is a faster growing yeast as compared to BCRC 21777, my procedure is as follows.
Procedure:
Prepare overnight culture at 30 degrees celsius at 150 rpm for 16 hours.
Use different ratios of inoculum for example, 1ml K-21 and 2 ml BCRC 21777, 1 ml K-21 and 4 ml BCRC 21777.
Place in an incubator and using HPLC to determine sugar consumption and agar plates for determining CFU/mL
I am sorry I have no experience doing co-culture and based on the papers I have read, co-culture is usually done in certain ratios. Really looking forward to your response.
