There's a problem with my mounting buffer, it is always liquid and always tends to enter under the cover slips which interferes negatively while imaging ...
Try vectashield H-1200/H-1400 its a hard mount and prevents any condensation from seepign below your coverslip, when u want to view them for a second time. Plus the H-1400 has DAPI. Hope this helps.
You can create an easy mounting medium yourseld, it remains "harder" than the commercial vectashield (on my experience). Is 80% glycerol in Pbs with 1% Propyl gallate. To prepare it mix before the propyl gallate in the pbs and then add the glycerol, to make completely homogeneus warm it (either in a water bath or a microwave). After that store it at +4°C covering it from the ligth. It will be very thick, I suggest you to use a cut 200microliter tip to put it on the slides, I use about 150 microliters)
Also, if you wish to do a nuclear staining you can add the Hoecst solution to this preparation diluted 1/10.000. For the moment I can say that this solution lasts at least 6 months if well preserved.
