Are any of you working on episomal MARS vectors?

I want to know how much time / generations they last WITHOUT an initial selection in mammalian cells (any type would do). I could clone a Kan gene into my vector for initial selection after the transfection but I don't want to.

Without an initial selection step in HEK's, for example, the MFI and the number of transfected cells drop significantly in my experiments.

Could anyone help me out?
