Liquid culture technique will definately help, you may use Middlebrook 7H9 or 7H11 for this purpose. If you have BD Bactect system accessible you may also use the MGIT tubes and the system. Innoculate the liquid culture tubes with your samples and incubate the tubes at 37 degrees celcius till you get desired growth. You may use suitable antibiotics in this medium to prevent the growth of contaminating microorganisms or my further purify the Mycobacterium bovis cultures.
