I just started working months ago with this peculiar cell line and I still cannot figure out the doubling time or passaging. Some times take 72h to see double amount of seeded cells, some times less, 48h. Are there some tips or secrets to handle this cell line?
I am using DMEM medium supplemented 10% Fetal bovine serum and 1% pen/strep. Passaging 1:2 or 1:3 maximum, once per week and using Trypsin 12 min for detachment.
The first question is how confident are you that you have Calu-3 cells? They should be fine being split at 1:4. Check for infections, including mycoplasma. Also check that your media bicarbonate and incubator CO2 match. 48 hrs is about the normal doubling time we see for Calu-3. Try changing your media to EMEM. good luck

Calu-3 cells were grown in flasks in complete Dulbecco’s Modified Eagle’s medium: F-12 containing 10% (v/v) fetal calf serum, 1% (v/v) nonessential amino acid solution, and 1% (v/v) L-glutamine solution and maintained.
