Hello, I tried several times to obtain two KO cell lines for ERN1 (IRE1 alpha) using a santa cruz crispr cas9 "double nickase". (HNSCC cancer cell lines : PECA-PJ41 and PECA-PJ34).

I tried different concentrations of lipofectamine / DNA / puromycin and each time the result is the same: all cells die after a few days of puromycin. I don't see any GFP signal at the fluorescence miscroscope.

Do you have any idea why the transfection goes wrong? Should I buy another type of crispr-cas9? Which alternative way do you recommend to obtain KO cell lines?
