I am doing MTT to calculate IC50 of Ptx using A549 cells. I prepare stock of Ptx (10mM) in DMSO but when i do further dilutions with culture media the Ptx separate out (settled down). Can any one advice me to overcome this problem?
You shoud dilute Ptx in DMSO or in solution of cyclodextrines. But first of all you need to elucidate action of it solvents to your model.
It will be better, if you will use as much as possible different solvents, as much as possible with maximal probability determine action of itself Ptx.
Dear Cergej, Thank you for your answer and i understand what you mean to say. I tried to reduce the concentration of DMSO in my working concentration as low as 0.1% and i also evaluated its toxicity by using DMSO control. But my real problem is the solubility of Ptx in the media. Advice me in this regard...
I ve never experienced such a king of problem. Did you check whether your drug is well stored( Humidity,Temperature...). Try to check the toxicity of DMSO.
