I found out that after isolating mitochondria from cells we first need to measure its protein concentration before any experiment . but can we directly measure mitochondria no. instated of that?
It is highly advisable to assess the protein concentration of your preparations (not just restricted to mitochondria) because you need it for normalization (e. g., when performing a Western Blot analysis you want to load equal amounts of protein per lane if you want to compare signals). You also do a loading control to check whether your samples are indeed present in equal amounts.
It is not possible to measure mitochondrial number definitively. Also not all mitochondria will be of same size. They are known to undergo fusion and fission as well. Thus it is highly advisable to measure mito protein conc to normalize your experiment results.
