I prepared PBMCs,s from human blood and differentiated them as immature dendritic cells in this process, monocytes adhere to the plates. What could be the reason for this phenotype?
Monocyte adhesion during dendritic cell differentiation could result from variations in cell culture conditions, such as coating of culture plates, or the duration of the differentiation process. Adjusting these factors may help minimize monocyte adhesion.
we isolated monocytes and cultured them under GM-CSF and IL4 to dendritic cells. We also observed that some cells were adherent and others were not. So on day 5 we examined both "fractions" once for the markers CD14, CD1a, CD80, CD86. There were no differences, so we assume that some cells are temporarily adherent and then detach again. There is always a proportion of adherent cells. We rinsed the cells on day 5 in order to reseed them for testing the test substances, which has the advantage that dead cells can also be removed.
