I have consulted multiple publication where Topoisomerase assays are performed using drug concentration of 50 and 100 micromolar?
why such high concentration used for this assay? Is there any probable reason?Why not researcher uses nanomolar drug concentration or less upto 10 micromolar concentration.

No there is no such any technical secret behind this. It is similar like other assays.. if you are confident that your compound is more potent and could show activity in nano molar concentration you could go with any concentration.
Secondly, to determine IC50 we have to plot a graph concentration vs inhibition for that also we have to take 3 or more than 3 concentrations. So testing on 25, 50 and 100 mico molar are generally chosen by the researchers. Selection of these concentrations are upto you...
