Can I ask you to provide more details on how you prepared your samples - protein concentration, lysis conditions, protein extraction, etc?

What is the target protein? Are you expecting it to be large?

What is the estimated molecular weight of the protein you are interested in? You may have to lower the acrylamide percentage further.

Dear Rokaya,

It seems the sample is having higher molecular weight proteins and obviously you have not mentioned clearly the desirable protein size of your choice. If protein size is more please go with 6% and 8% gel. Compare the bands clarity and resolution after staining.
