Currently planning fungal culture for later epigenetic experiments, and was debating whether to use minimal media or the typical Sabouraud.
I was wondering, however, why Sabouraud agar seems to come out in a relatively wide range of colours - from pale yellow to amber brown - and whether this apparent difference might be enough to cause effects at the genetic level.
Does anybody know the cause of this colour difference from batch to batch?
To the best of my knowledge it id because of the heating effect caused during autoclaving. If your autoclaving cycle time is not consistent. Alternatively it could also be due to use of an old bottle of sugar that you are adding to the sab. agar. Try with another sugar bottle.
Sabouraud agar has many modifications in medical mycology. The colour depends on the constituents of the medium. The constituents may vary depending on the suppliers, age of the constituents, method used in the preparation of the medium. Age of the prepared medium and time period in storage.
Millard reaction is primarily the reason for the browning of the medium.
Age of Constituents added Prolonged heating and cooling cycles can enhance the color (Yellow to Brown).
Mellentoin like compound is generated during the Millard reaction (commonly seen in Sugar manufacturing).
Solution: Seperatly autoclave protein (Peptone and/or Yeastextract) and add to other ingradients admix and heat in a water bath for a few min and prepare plates.
The color consistency will remain the same every time.
