Hi guys!

I have a problem with my pKD46(which has heat-sensitive origin pSC101) not cured, after lambda red recombination. I confirmed my recombinant bacmid right after red recombination by PCR and it was successful. However whenever i tried to cure pKD46(which is not necessary after the reaction), it does not get off..

pKD46, which i'm using has gentamicin resistance gene. I cured my recombinant at 42℃ for 48 hours and checked by streaking on (KanR+ChloramR+GenR) and (KanR+ChloramR) LB agar plate respectively. (Recombinant bacmid has KanR and ChloramR and its in the DH10B).

what seems to be a problem ??

+ I heard, successive subcultures and incubation at 43℃ helps pKD46 curing.. and if this is true.. how long should i give successive subcultures? please help if you guys have good tips for plasmid curing. Thank you for reading :)
