I am using 4 gel units of MNase enzyme for 2ug of polynucleosomes in expectation to get mononucleosomes. I kept this reaction at 37degC for 15 minutes. This reaction worked two times, confirmed by checking the DNA fragment on a native PAGE representing the 147bp wrapped DNA. The same reaction is not working anymore. Why could that be happening?

Achim Recktenwald - Hey Achim, yes my enzyme is still working as I perform lambda DNA digestion with it and it works.

Did you make any changes in how you obtain your sample and, specifically, your sample is prepared?
Do you use a new lot bottle of a chemical? Even a different lot?
Do you use different equipment? Has the equipment changed, e.g., been serviced, calibrated, etc?
