I am planning to introduce a point mutation in Saccharomyces cerevisiae, strain background CEN.PK. So, I was curious in asking whether instead of adding mutation using primers, joining mutated version of the gene with a selectable marker and then transforming in yeast, or performing CRISPR, is there any other way of doing the same, maybe more efficient, less time-consuming and reliable of course?
