There's this problem I have been getting while separating the serum from blood collected from mice by terminal bleeding from heart. I collect 0.5-1.0ml blood, keep it in ice until centrifuguation at 8000rpm for 5min on table top centrifuge. In some of the tubes, the serum is completely jelly like while in others it is completely fine.
Has anyone been through similar problems? What could be the possible explanation for this observation?
I also have the same problem with peritoneal lavage collected with 2ml of normal saline.

if you need serum then do not keep blood in Ice soon after collection. first let it coagulate and centrifuge to recover serum then you can keep in cold. try to collect blood before mice is feeded.
In case after feeding some time serum give turbid and jelly appearance what i have seen.
The jelly like appearance is due to clotting. You can prevent this by collecting your blood in heparin tubes. For peritoneal fluid, collect in saline containing heparin. Use the concentration of heparin recommended by supplier as anti-coagulant.
I have had to troubleshoot through a similar issue. As James pointed out, you need to let the blood coagulate for 20-30 minutes at room temperature. You want the blood to fully coagulate to remove as many clotting factors as possible (fibrinogen, etc.). Don't leave it out for more than 30 minutes and you shouldn't have problems with protein degradation. If that still doesn't work, you can also increase the g forces to pull more fibrous material out. I am facing same issue with my sample collected from patients with Hepatic enceph in ACLF.
On separating plasma or serum jelly like consistency formed.
All patient are cirrhotic, bleeding.
I even tried after half an hour same thing happen.
Can any one predict the reason.
