there is no need to do what you describe, that way you are also loosing the possibility of loading more samples per gel. It is however recommended to not leave empty wells as the lines next to them will tend to expand. It could also produce a non even electrophoresis. For these reasons people normally add loading dye to empty wells.

Hi, if your lanes are empty its recommended to add cracking buffer to prevent edge effects.

We usually add SDS-PAGE sample buffer (1X) in all empty wells to avoid diffusing the bands from neighboring wells.
