I try to load DNA plasmid in Wheel gel electrophoration, but the DNA cannot load directly and brustle. is it any correlation with ethanol remaining in my DNA plasmid after isolastion?
If your DNA is not dry traces of ethanol remain. Ethanol contamination can lead to problems with repulsion of DNA when loading agarose gels electroporation etc. Thus remove all traces of ethanol and alllow plasmid pellet to dry at room temp for 15 min before resuspending in aqueous buffer
