I coated PES/PVP membranes with 1mg/ml and 200ug/ml human fibronectin (30min, 37C) after which HUVECs were seeded on top. I try to improve attachment to the membrane and have found numerous examples in literature where they use fibronectin to increase the HUVEC attachment. I did find an increase from 100 +-46.2 cells/cm2 (uncoated control) to 129 +- 20.0 cells/cm2 (coated). Concerning the huge standard deviations this increase is to small to be significant.

Does anyone have an idea of why the positive effect of fibronectin might be so small?
