I am trying to detect ND-6 protein expression in gastric cancer tissue sample and cell line. I have tried Anti-ND6 antibody AV50255-50UG from sigma and it had not given me any specific band which abcam suggested at around 25 kDa. I have tried more than 3 times replication but i always got same results.
Appreciate, if any one can provide feedback with their experience/expertise with the ND-6 western blot analysis.

For proper answer details of procedure to make homogenization of the tissue and the subsequent centrifugation steps are needed.
Also a positive controls should be used.
Second the need for a positive control as mentioned by @Rafael. Without this you are working blind.
