I tested 1 compound and 5-ASA (as positive control) in the DSS-induced colitis mice model.
Neither my compound nor 5-ASA showed anti-inflammatory activity.
In fact, 5-ASA was a bit pro-inflammatory.
How can I justify the slight pro-inflammatory activity of 5-ASA? What can be the reasons?

Would be great if you can provide some more background about your model (DSS concentration, duration), dose/application route of 5-ASA. How did you check inflammatory activity?
5-ASA (75 mg/kg/day, oral) for 8 days, day 1 to day 8.
DSS (5 % in drinking water) for 6 days (day 1 to day 6).
Male C57BL/6 mice were used.
mRNA and Protein levels of TNF-a, MCP-1, and IL-6 were analyzed in colons.

For male C57BL/6(J?), 5% DSS for 6 days might be a pretty severe treatments. Probably a large number of animals will die.
At our institution, we used 2-2.5% DSS treatment because of the issue of the mortality rate and weight loss. 5-ASA (CMC-Na suspension) alleviated the symptoms of colitis and inflammation in colon mucosa, and downregulated the expression of pro-inflammatory cytokine genes. The other treatments themselves were almost the same as yours.
