i brought stigmasterol for Sigma but it only dissolve in chloroform ..........although i have tried it to dissolve in DMSO but it did not dissolve in DMSO ....so i have tried to dissolve it first in chloroform then DMSO then it dissolve but its contain more than 1% chloroform which is more than 0.1% and lethal for neuronal cell so is there any specific solvent to dissolve it .......... is that tween is ok for cell culture
Other than chloroform stigmasterol should be soluble in diethyl ether, acetone and benzene. As an "equivalent" to chloroform you could try ethyl acetate. Stigmasterol will not dissolve in strongly polar solvents (solvents with a high dipole moment). Ethanol is too polar (too water like) and, while aprotic, so is DMSO (which is even more polar the ethanol or water).
Incidentally, the most commonly used solvent in chemistry is water.

And prof Wolfram Meier-Augenstein i was little bit concern about as you said, theoriticaly stigmasterol shoud not dissove in ethanol according to polarity and i am also thinking like that so i dont try to dissolve it in ethanol first but i see that stigmasterol dissolve in ethanol why if you kindly explain this matter it would be great pleasure for me
It is good to know that Ethanol can work for you. As Ethanol itself is relatively low toxic compared to other organic solvents.
I am not a chemist. I worked on lots of odd organic extracts including natural extracts and their modified products, for drug screening on human cells. DMSO and Ethanol are usual choices for those odd compounds. Of course, water is far more commonly used in non organic world.
Dear Md Nazmul Haque Habib. Virtually all organic compounds are soluble to a larger or lesser degree in one or more organic solvents. Only when its solubility is < 1mg/mL is a compound deemed insoluble. Solubility of stigmasterol in ethanol is just above this threshold with a solubility of about 2mg/mL. However, compared this to its solubility in chloroform of about 50mg/mL and I am sure you will understand my comment of stigmasterol being hardly soluble in ethanol. That being said, I appreciate why one would prefer ethanol over chloroform or indeed benzene even if solubility is limited.
Stigmastrol is soluble in most of the organic solvents viz., acetone, benzene, diethyl ether etc.
