Constructs with long (300-600 bp per repeat) inverted repeats with appropriate intron in between usually gives the best results. There is a number of vectors that can be used for this purpose.
The factors to be considered for the construct design are: minimizing the off-target effects by selecting a specific sequence, and (optional) looking for a region with high number of predicted efficient siRNA.
In filamentous fungus Mucor circinelloides, the efficiencies of gene silencing when transformed by the inverted repeat sequence and a single of sequence are not significant difference.
