Hi!

Does anyone knows what type of ladder I sould use for circular mtDNA in an agarose gel?

I am trying to isolate mammalian mitochondrial DNA from nuclear DNA using a 0.5% agarose gel, following a chelex extraction of both nuclear and mt DNA. Since the mtDNA is not very concentrated at this point, bands are not visible, so I need a ladder to find the 16.5kbp height. But circular DNA moves faster than the linear one... so, is there a ladder out there made for mtDNA? Or plasmids?

I've been looking for info, but it seems isolation of complete, unpurified mtDNA on a gel is not that common ;)

Thank you!
